Concentrate and purify AAV in under 4 hours.

Achieve high yields & scale-up linearly with minimal optimization.

IsoTag™ AAV Reagent

Streamlined Adeno-associated Virus Purification without Chromatography

IsoTag™ AAV is a specialized reagent, engineered for the challenging requirements of small and large-scale manufacturing. It enables a robust, efficient, and consistent downstream purification process for a broad spectrum of adeno-associated virus (AAV) serotypes. Purification with IsoTag AAV does not require the use of chromatography processes nor column packing, and is designed to reduce the costs and challenges associated with scaling from R&D to commercial production.

Bind in solution

The binding domain targets a specific biomolecule of interest, binding in seconds (no mass transfer limits)

Concentrate & wash

Pull target into large droplets that are in a separate phase from other aqueous contaminants.

Final product

Efficiently collect high-yield product using existing equipment and off-the-shelf consumables.

Results from the centrifugation (ALPS-CF) buffer screen directionally translate to the scalable tangential flow filtration (ALPS-TFF) process. IsoTag™ AAV Reagent is paired with platform processes that use existing equipment and off-the-shelf consumables.

ALPS-CF small scale: 1mL to 50mL

Use with microcentrifuge tube, 50mL tube and beyond with low-speed (5000 x g) centrifuge

ALPS-TFF scalable: volumes > 1L

Use with existing TFF equipment and hollow fiber filters

+ Scalability

ALPS-TFF scales linearly, increasing filter surface area with harvest volume

+ Yield

>90% capture and 75% elution of both total virus capsids

+ Quality

≥4 LRV of contaminating host cell proteins & DNA in harsh conditions

IsoTag™ reagents are paired with platform processes that use existing equipment and off-the-shelf consumables to achieve high purity and yield in hours. IsoTag AAV can utilize a microfiltration hollow fiber and, as such, is run in permeate control mode to maintain a constant flux throughout the process. This allows completion of the process in under 4 hours regardless of scale & can consolidate multiple unit operations, into one-step TFF.

Improved yields

High product quality

No size target limitations

A variety of AAV serotypes demonstrated binding efficacy in small-scale capture tests
(note: elution may require optimization)
